Increased Resistance to Oxidative Stress by Cloned Mn-SOD Gene in Escherichia coli

Abstract

|
The manganese superoxide dismutase(Mn-SOD) is a metalloenzyme which plays an important role in protecting cells against oxygen toxicity. The cloned Mn-SOD gene of Escherichia coli was subcloned into the E. coli-Lactococcus spp. shuttle vector pGKV21 to make the plasmid pGKV-SOD. The pGKV-SOD was then transformed into the E. coli strain HB101 by electroporation. SOD activity of the resulting transformant was 26.5% higher than that of E. coli harboring pGKV21 vector itself. In line with this result, the E. coli transformant showed the increased resistance to paraquat-mediated inhibition of growth. Paraquat is known as a promoter to the formation of a superoxide radical anion, O^£þ_2Based on these data, we can conclude that the cloned Mn-SOD provides protection in E. coli against oxygen toxicity.
